About the Lab 

We investigate the genes and proteins involved in biosynthesis of complex polysaccharides and glycoproteins in the plant cell wall, using arabidopsis and rice as model plants. Besides the fundamental scientific importance of these studies, understanding wall biosynthesis and regulation may allow us to develop crop plants that are more suitable feedstocks for biofuel production. Techniques used in the lab include biochemistry, spectroscopy, molecular genetics, cell biology and bioinformatics. ResearchBiosynthesis of the plant cell wall, glycosyltransferases, plant biotechnology.
 Polysaccharides constitute the major part of plant cell walls. The cell wall polysaccharides play important roles in maintaining shape and strength of plants, and for interactions with the environment, including pathogens and symbiotic organisms such as mycorrhizal fungi. There are many different types of polysaccharides in the wall and a large number of enzymes are needed for their biosynthesis. The rational development of biofuel crops with improved properties will require a much better understanding of cell wall biosynthesis and the role of individual polymers for plant growth and development. In the Cell Wall Biosynthesis Group we aim to:

 

  • Identify the glycosyltransferases and accessory enzymes required for biosynthesis of matrix polysaccharides and glycoproteins in the cell wall.


  • Develop efficient methods for biochemical analysis of cell wall biosynthetic enzymes

.
  • Understand the role of specific cell wall polysaccharides for growth and development, interactions with microorganisms, and abiotic stress.
  • Discover the regulatory and signal transduction mechanisms used by plants to dynamically control cell wall composition.
  • Use the knowledge about cell wall biosynthesis to make targeted modifications of cell wall composition in transgenic plants and determine the effects on plant growth and biofuel yield.
  • Participate in forward genetic screens for plants with improved properties for biofuel production.



The polysaccharides in plant cell walls are often grouped into cellulose, hemicellulose and pectin. The major hemicellulose in grasses and in secondary walls is xylan, which is synthesized from UDP-xylose, UDP-arabinose and UDP-glucuronic acid by the action of glycosyltransferases in the Golgi vesicles, which add sugar residues onto existing acceptor molecules, which may be oligo- or polysaccharides. Xylans can be acetylated and cereal arabinoxylans also contain esterlinked ferulic and p-coumaric acid the biosynthesis of which is not understood. The acetyl and feruloyl esters are a major challenge for biofuel production. In order to characterize the individual glycosyltransferases and other biosynthetic enzymes it is essential to develop assays that are dependent on heterologously expressed enzymes and exogenous acceptor molecules.Pectin is a very complex polysaccharide consisting of many different sugars and glycosidic linkages. More than 60 transferase enzymes are believed to be required for pectin biosynthesis but only four different enzymes have been identified and characterized.Xylan is the most abundant matrix polysaccharide in biomass. A large number of enzymes and transporters are required for xylan biosynthesis, but many of these proteins have not been identified and their interplay and regulation is not understood. Environmental interactionsSince biofuel crops are likely to be grown on marginal lands, it is essential that they will be able to grow well under stressful conditions and with low inputs, especially water and fertilizers.
We are interested in tolerance to drought stress and to temperature stress, and we investigate interactions between plants and mycorrhizal fungi, which are important for nutrient and water uptake. We are especially interested in understanding the role of plant cell walls in these interactions.

Plant Biotechnology

Many of the strategies used to modify plant cell walls can also be applied to modify other metabolic pathways. Currently, we investigate methods to develop plants with increased oil yield.
